Introduction –
Cells in cell culture respond sensitivity to infection mostly by the virus. so require to them store at very low temperature for further use for study or gain to the desired product. for this use many methods like frizzed to them at -85ºC deep freeze and cryopreservation at 196ºC in liquid nitrogen etc. or more. but storage to liquid nitrogen increase in the cost of this.
CONTAMINATION IN ANIMAL CELL CULTURE
Preservation–
It means have to animal cells at a very low temperature called preservation. in case of cryopreservation, all ampoule of cells has in liquid nitrogen at -196ºC for a long time. but in this method, a major disadvantage is the formation of ice crystals inside cells during deep freezing so use DMSO ( dimethyl sulfoxide) to protect to cells by mechanical damage created by ice formation.importance to preserve cells to maintain the lowest number of passage for a long period. during handling to cells at very low temperature in liquid nitrogen cause ice burn to cells.
one major benefit to preserve the cells at very low temperature is so we can be isolated to harmful virus-like adeno and herpes simplex virus. that we can be confirmed by the use of polymers chin reaction. in process of freezing’s to cells temperature reduce slowly to protect to cells by damage about 30ºC to 60ºC than again increasing in temperature for long time storage.
